Analysis of Drugs in Blood Plasma by Co-Sense for BA Biological Sample Analysis System

The Co-Sense for BA biological sample analysis system incorporates a unique online dilution bypass and column-switching technology using the newly developed Shim-pack MAYI-ODS pretreatment column. It automates the deproteinization and other pretreatment operations required for biological samples.

As drugs in blood plasma are often bound to proteins, deproteinization can result in reduced drug recovery rates.
Consequently, when using the Co-Sense for BA system, it is important to select a mobile phase for sample injection that promotes dissociation of the drug from the protein. As the drug and protein bind due to ionic interactions and hydrophobic interactions, the control of the pH and ionic strength of the mobile phase for sample injection and the addition of an organic solvent can be effective. In this example, the mobile phase for sample injection was acetic acid (sodium) buffer solution or acetic acid (sodium) buffer solution spiked with 10 % max. acetonitrile. The injected blood plasma sample was automatically diluted in the mobile phase by a factor of eight in the online dilution bypass.

The analysis of eight drug components in blood plasma is shown below. The sample was created by spiking the prefiltered human blood plasma with drug standards.
The Shim-pack MAYI-ODS pretreatment column selectively eliminated proteins and other macromolecules, while effectively trapping low-molecular compounds, such as drugs.

HPLC is widely used for the analysis of drugs in biological samples. However, sample pretreatment is a more important factor in achieving the targeted accuracy and sensitivity than the chromatography conditions. Co-Sense for BA consists of a unique hardware configuration incorporating an online dilution bypass and the newly developed Shim-pack MAYI-ODS on-line SPE column for seamless automatic operation from sample pretreatment to analysis. Online treatment of blood serum and blood plasma samples offers higher analysis efficiency and labor savings.
